Role of orFD Pseudomonas aeruginosa H103 Gene in Glucose Uptake
Authors
Abstract:
Background:Pseudomonas aeruginosa is a gram negative non facultative bacterium and one of the members of normal flora in different sites of body in healthy humans.this bacterium can resist in fluids and hospital environments for a long time.Pseudomonas aeruginosa has two systems for glucose uptake:a low affinity oxidative pathway and a high affinity phosohorylative pathway.Although the role of these genes are unknown by now, they may have a role in regulation of glucose uptake. In the present study, the role of orFD gene in glucose uptake in P.aeruginosa has been investigated. Method:orFD fragment were cloned in pUCP20 as vector and the recombinant plasmid transferred into WMA200 strain of P.aeruginose,a mutant strain of p.aeruginosa with a chromosomal deletion of orFBCD.So we compared the rats of glucose uptake by p.aeruginosa by using labeled glucose under condifions at low substrate concentration and low cell density. Results:Carbohydrate uptake patterns differed considerably among three strains.the wild type is able to uptake glucose at a faster rate than the mutant:however, the mutant complemented with orFD shows an intermediate uptake comparing to the wild type and the mutant. Conclusion:orFD gene has an important role in carbohydrate uptake in p.aeruhinosa strains however fuether studies are required to determine the involved mechanism.

Role of PA3574 (nalD) gene in development of ciprofloxacin resistance in Pseudomonas aeruginosa isolates
Background & Aims: Pseudomonas aeruginosa is a gram-negative opportunistic pathogen and one of the mortality causes of nosocomial infections. One of the drug resistance mechanisms in P.aeruginosa is mutation in negative regulator genes of mexAB-oprM efflux pump system such as nalD. The aim of this study was to investigate the role of nalD mutations in P. aeruginosa isolates of Guilan province i...
full textOuter Membrane Protein D Gene in Clinical Isolates of Pseudomonas Aeruginosa and its Role in Antibiotic Resistance
Background & Objectives: Pseudomonas aeruginosa is a common cause of nosocomial infection. OprD protein is a specific protein regulating the uptake of carbapenem antibiotic. Loss of OprD is the main mechanism of Pseudomonas Aeruginosa resistance to carbapenem. In this study, the presence of OprD gene is investigated in isolated Pseudomonas Aeruginosa in burn patients of Ghotboddin hospital in S...

full textRole of the novel OprD family of porins in nutrient uptake in Pseudomonas aeruginosa.
To circumvent the permeability barrier of its outer membrane, Pseudomonas aeruginosa has evolved a series of specific porins. These channels have binding sites for related classes of molecules that facilitate uptake under nutrient-limited conditions. Here, we report on the identification of a 19-member family of porins similar to the basic-amino-acid-specific porin OprD. The members of this fam...
full textRole of apoptosis in Pseudomonas aeruginosa pneumonia.
Apoptosis plays a central role in the complex balance between invading pathogen and host defense. Depending on the pathogen, apoptosis of host cells may be beneficial or detrimental to survival. Studies using a clinically relevant intra-abdominal model of sepsis (peritonitis) have indicated that two types of cells, lymphocytes and gastrointestinal epithelial cells, undergo accelerated apoptosis...
full textOxygen regulation of nitrate uptake in denitrifying Pseudomonas aeruginosa.
Oxygen had an immediate and reversible inhibitory effect on nitrate respiration by denitrifying cultures of Pseudomonas aeruginosa. Inhibition of nitrate utilization by oxygen appeared to be at the level of nitrate uptake, since nitrate reduction to nitrite in cell extracts was not affected by oxygen. The degree of oxygen inhibition was dependent on the concentration of oxygen, and increasing n...
